[연구내용]
생명체는 다양한 기능을 수행하는 세포라는 단위로 구성되어 있으며, 이들은 유전자들에 저장된 정보의 발현 산물인 단백질들의 조합으로 이루어져 있다. 생명현상의 유지를 위해 세포들간의 정보교환이 이루어져야 하는데 이들은 신호전달 (Signal Transduction)이라는 과정을 통해 형성된다. 이러한 일련의 신호전달과정은 단백질 분자간의 네트웍이 조절됨으로서 가능하며 이 과정의 이상은 암, 당뇨, 치매, 불임등과 같은 질병들의 직접 또는 간접적인 원인이 된다. 따라서 유전자 수준에서의 연구뿐만 아니라 단백질의 변형이나 단백질 결합 복합체의 변화에 대한 연구방법이 요구된다.
우리 분자신호전달 연구실에서는 암세포, 역분화/줄기세포 (인간과 마우스) 및 임파구 신호전달체계에서 단백질분해과정 및 세포의 성장과 분화를 조절할 것으로 생각되는 단백질분해조절 효소 (Deubiquitinating enzyme: 탈유비퀴틴화 효소)의 생물학적 기전을 규명하고 있다. 최근 이들 세포에서 새로운 탈유비퀴틴화 효소 유전자들을 발굴했으며, 이들의 생물학적 기능을 이해하기 위해 분자유전학 기법을 이용하여 연구하고 있다. 이 프로젝트의 목표는 암세포, 줄기세포, 및 임파구의 성장과 분화를 조절할 수 있는 새로운 방법을 찾는 것이다.
두 번째 연구영역으로는 습관성 유산 및 다낭성 난소증후군을 유발하는 유전자 및 유발요소를 찾고 분자적 기전과 역할을 규명함으로써 궁극적으로는 세포 및 유전자치료를 이용하는 임상적 응용에 연구의 초점을 맞추고 있다. 특히 임신의 과정은 빠른 유전자 발현 양상의 변화가 요구하므로 신호전달계에서의 조절이 잘못 될 경우 바로 태아성장의 실패와 연결된다는 점에서 그 중요성이 더욱 강조되고 있다. 본 연구실에서는 습관성 유산질환 및 다낭성 난소증후군 관련 유전체 및 단백체를 찾기위해 정상과 질환 환자로부터 얻은 샘플로부터 cDNA subtraction method, DD-PCR, 및 2DE-PAGE/MALDI-TOF/MS 분석등을 이용하여 정상인과 환자와의 유전자 발현의 차이를 비교함으로서 습관성 유산질환 및 다낭성 난소증후군의 진단과 치료의 방법을 개발하고자 한다.
